RESUMO

PURPOSE: To investigate the prevalence of lower urinary tract symptoms/benign prostatic hyperplasia in a Korean population. MATERIALS AND METHODS: The Korean Prostate & Voiding Health Association provided free prostate-related community health care and conducted surveys in all regions of Korea from 2001 to 2022 with the cooperation of local government public health centers. A total of 72,068 males older than 50 were surveyed and analyzed. History taking, International Prostate Symptom Score (IPSS), transrectal ultrasonography, prostate-specific antigen (PSA) testing, uroflowmetry, and urine volume testing were performed. RESULTS: The mean prostate volumes in males in their 50s, 60s, 70s, and 80s or above were 24.7 g, 27.7 g, 31 g, and 33.7 g, respectively. The proportion of males with high PSA greater than 3 ng/mL was 3.8% among males in their 50s, 7.7% among males in their 60s, 13.1% among males in their 70s, and 17.9% among males 80 years of age or older. The mean IPSS total scores in males in their 50s, 60s, 70s, and 80s or above were 10.7, 12.7, 14.5, and 16, respectively. Severe symptoms were reported by 27.3% of males, whereas 51.7% reported moderate symptoms. The mean Qmax in males in their 50s, 60s, 70s, and 80s or above were 20 mL/s, 17.4 mL/s, 15.4 mL/s, and 13.8 mL/s, respectively. CONCLUSIONS: In this population-based study, mean prostate volume, IPSS, PSA, and Qmax were 30.6±15.1 g, 14.8±8.2, 1.9±4.7 ng/mL, and 15.6±6.5 mL/s, respectively. Aging was significantly associated with increased prostate volume, PSA levels, and IPSS scores, and with decreased Qmax and urine volume.

Cutting-edge nanoelectrokinetic technology in this work provides a breakthrough for the present clinical demands of molecular diagnosis to detect a trace amount of oncogenic mutation of DNA in a short time without an erroneous PCR procedure. In this work, we combined the sequence-specific labeling scheme of CRISPR/dCas9 and ion concentration polarization (ICP) mechanism to separately preconcentrate target DNA molecules for rapid detection. Using the mobility shift caused by dCas9's specific binding to the mutant, the mutated DNA and normal DNA were distinguished in the microchip. Based on this technique, we successfully demonstrated the dCas9-mediated 1-min detection of single base substitution (SBS) in EGFR DNA, a carcinogenesis indicator. Moreover, the presence/absence of target DNA was identified at a glance like a commercial pregnancy test kit (two lines for positive and one line for negative) by the distinct preconcentration mechanisms of ICP, even at the 0.1% concentration of the target mutant.

We explored the physiological effects of inhaling basil essential oil (BEO) and/or linalool and identified odor-active aroma compounds in BEO using gas chromatography/mass spectrometry (GC-MS) and GC-olfactometry (GC-O). Linalool was identified as the major volatile compound in BEO. Three groups of rats were administered BEO and linalool via inhalation, while rats in the control group were not. Inhalation of BEO for 20 min only reduced the total weight gain (190.67 ± 2.52 g) and increased the forced swimming time (47.33 ± 14.84 s) compared with the control group (219.67 ± 2.08 g, 8.33 ± 5.13 s). Inhalation of BEO for 5 min (392 ± 21 beats/min) only reduced the pulse compared with the control group (420 ± 19 beats/min). Inhalation of linalool only reduced the weight of white adipose tissue (5.75 ± 0.61 g). The levels of stress-related hormones were not significantly different among the groups. The total cholesterol and triglyceride levels decreased after inhalation of BEO for 20 min (by more than -10% and -15%, respectively). Low-density lipoprotein cholesterol levels were lowered (by more than -10%) by the inhalation of BEO and linalool, regardless of the inhalation time. In particular, BEO inhalation for 20 min was associated with the lowest level of low-density lipoprotein cholesterol (53.94 ± 2.72 mg/dL). High-density lipoprotein cholesterol levels increased after inhalation of BEO (by more than +15%). The atherogenic index and cardiac risk factors were suppressed by BEO inhalation. Animals exposed to BEO and linalool had no significant differences in hepatotoxicity. These data suggest that the inhalation of BEO and linalool may ameliorate cardiovascular and lipid dysfunctions. These effects should be explored further for clinical applications.

This study investigated chemosensory degradations of soybean and canola oils with repeated frying in order to estimate the quality of the oils. METHODS: Chemical parameters including oxygen induction time, acid value, p-anisidine value, malondialdehyde, and total polar compounds were measured. Electronic nose and electronic tongue analyses were performed to assess sensory properties. Multivariate analyses were employed to investigate relationships among tastes and volatile compounds using principal component analysis (PCA) and Pearson's correlation analysis. RESULTS: All chemical parameters increased with repeated frying in both oils. Electronic nose analysis found ethyl butyrate, 2-heptenal, and 2,4-pentanedione as major volatiles for soybean oil and ethyl butyrate and linalool for canola oil. As the numbers of frying increased, all volatiles showed an increased concentration in various extents. In multivariate analyses, ethyl butyrate revealed strong positive correlations with sourness, umami, and sweetness, and umami showed strong positive correlations with sourness and saltiness (p < 0.05). PCA confirmed that in PC1 with 49% variance, sourness, saltiness, and umami were at similar rates while acetyl pyrazine, 2,4-pentadieone, and 1-octanol were found at similar rates. Canola oil was chemically more stable and less susceptible to deterioration in all chemical parameters compared to soybean oil, resulting in a relatively better quality oil when repeatedly fried. CONCLUSION: The results suggested that minimum repeated frying (5 times) degrades chemosensory characteristics of both oils, thereby compromising their quality. The findings of this study will be utilized as a foundation for quality control of fried foods in food industry, fried food development, and fast-food industry.

Splendid advancement of micro/nanofluidic researches in the field of bio- and chemical-analysis enables various ubiquitous applications such as bio-medical diagnostics and environmental monitoring, etc. In such devices, nanostructures are the essential elements so that the nanofabrication methods have been major issues since the last couple of decades. However, most of nanofabrication methods are sophisticated and expensive due to the requirement of high-class cleanroom facilities, while low-cost and biocompatible materials have been already introduced in the microfluidic platforms. Thus, an off-the-shelf and biodegradable material for those nanostructures can complete the concept of an eco-friendly micro/nanofluidic platform. In this work, biodegradable materials originated from well-known organisms such as human nail plate and denatured hen egg (albumen and yolk) were rigorously investigated as a perm-selective nanoporous membrane. A simple micro/nanofluidic device integrated with such materials was fabricated to demonstrate nanofluidic phenomena. These distinctive evidences (the visualization of ion concentration polarization phenomenon, ohmic/limiting/over-limiting current behavior and surface charge-governed conductance) can fulfill the requirements of functional nanostructures for the nanofluidic applications. Therefore, while these materials were less robust than nano-lithographically fabricated structures, bio-oriented perm-selective materials would be utilized as a one of key elements of the biodegradable and eco friendly micro/nanofluidic applications.

Over the past several decades, separation and preconcentration methods of (bio)molecules have been actively developed for various biomedical and chemical processes such as disease diagnostics, point of care test and environmental monitoring. Among the great developments of the electrokinetic method in a micro/nanofluidic platform is the ion concentration polarization (ICP) phenomenon, in which a target molecule is accumulated near a permselective nanoporous membrane under an applied electric field. ICP method has been actively studied due to its easy implementation and high preconcentration/separation efficiency. However, the dynamic behavior of preconcentrated analytes has not yet been fully studied, especially driftless migration, where the applied electric field is orthogonal to the direction of the drift migration. Here, we demonstrate anomalous shapes of preconcentrated analytes (either plug or dumbbell shape) and the morphologies were analytically modeled by the leverage of convection and diffusion migration. This model was experimentally verified with various lengths of DNA and the limiting cases (convection-free environment in paper-based microfluidic device and extremely low diffusivity of red blood cells) were also shown to confirm the model. Thus, this study not only provides an insight into the fundamental electrokinetic dynamics of molecules in an ICP platform but also plays a guiding role for the design of a nanofluidic preconcentrator for a lab on a chip application.

RESUMO

A 39-year-old female was suffering from cold-induced Raynaud's attacks in both hands and feet, with symptoms being most severe in her left foot. The patient did not respond to medical treatments and was referred to our department of pain medicine. We performed sequential bipolar radiofrequency lumbar sympathectomy to the patient, which offered a long duration of symptom relief. Sequential bipolar radiofrequency lesions could create continuous strip lesion, and thus, could achieve better results, while the potential risk of liquid neurolytic agents could be avoided.
